Yes, E=mc^2 is the ratio at which they are converted into each other. We use energy to produce matter in particle accelerators - it's how we synthesise heavy elements and it's how we discover new fundamental particles (like the higgs boson).

I have no clue why the last guy to answer this put a load of stuff about the big bang, abiogenesis and quark gluon plasma up.
